Sample Requirements for DNA Profiling & Cell Culture QC

Use the overview below to prepare samples for Fragment Length Analysis, Cell Line Authentication and Mycoplasma Testing.

Fragment Length Analysis

Samples for Fragment Length Analysis must be submitted in 96-well plates. Individual tubes cannot be processed.

Requirements:

  • ≥10 µl diluted, labeled PCR product per well
  • Plate must be labelled with a Microsynth FLA barcode plate label
  • Ready-to-load samples must already contain size standard / Hi-Di formamide mix
  • Empty wells must be filled with distilled water
  • Plates must be sealed properly for safe shipment

RCT option: Dye sets using the RCT signal setting use the same size standards as the corresponding standard dye sets.

Cell Line Authentication

For Cell Line Authentication, submit either a cell pellet or isolated genomic DNA.

Accepted sample types:

  • Cell pellet: 1–5 million cells, washed twice in PBS or another appropriate buffer, resuspended in 0.5 ml of 70–90% ethanol
  • Isolated DNA: ≥50 µl genomic DNA at 50 ng/µl in Tris or low-EDTA buffer

Mycoplasma Testing

For Mycoplasma Testing, collect cell culture supernatant from cultures grown for at least 72 hours without medium change. Cell cultures should be approximately 80–90% confluent, but not overgrown.

Sample preparation overview:

  • Collect cell-free culture supernatant
  • Centrifuge to remove cells and debris
  • Transfer 150 µl supernatant into a 1.5 ml or 2 ml flip-cap tube
  • Heat-inactivate at 95°C for 10 minutes
  • Label the tube with a Microsynth Mycoplasma barcode label
